Such new initiatives as integrated continuous bioprocessing are inspiring new intensified manufacturing approaches including high-cell density perfusion culture. These culture formats can change basic process parameters resulting in altered cycle rates and metabolic demands by the cultured cells. This can place unique demands upon the culture media.
However, the fed-batch mode of operation is still the most employed, and work continues in the optimization of its materials and protocols. This work begins with the selection of a robust chemically-defined basal medium and feed formulation. Studies in clone and process-optimized feed protocols as well as appropriate formulation adjustments result in optimized volumetric productivity and more consistant product quality. Presented here are case studies on culture media and feed development towards both perfusion and fed-batch processes.
